P53 is a tumor suppressor protein; whereas, proliferative cell nuclear antigen (PCNA) is a proliferative marker. The aim of this study was to evaluate the immunohistochemical expressions of P53 and PCNA in oral leukoplakia with and without dysplasia. Materials & Methods: The expression of P53 and PCNA proteins were determined immunohistochemically in paraffin sections of 53 leukoplakias (17 without dyplasia, 15 with mild dysplasia, 14 with moderate dysplasia and 7 with severe dysplasia), in addition to 10 normal oral mucosa. Statistical analysis was performed using One-Way ANOVA, Kruskal-Wallis, and spearman tests. Results: P53 protein was expressed in 60% of normal oral mucosa, 82.4% of leukoplakias without dysplasia and 100% of leukoplakias with dysplasia. PCNA protein was expressed in all of the specimens. There were statistical differences between P53 and PCNA expression among normal mucosa, leukoplakias with and without dysplasia, regarding their grading and severity (P<0.001). There was a positive correlation between P53 and PCNA expression. Conclusion: Increase in P53 and PCNA expression is related to the degree of dysplasia in oral leukoplakias. Therefore, we can use these markers in order to indicate and grade dysplastic lesions more exactly. P53 پروتئین مهارکننده رشد تومور و PCNA بیومارکر تکثیر سلولی است. هدف از این مطالعه تعیین و مقایسه بروز آنتی ژن P53 و PCNA در لکوپلاکیای مخاط دهان با و بدون دیسپلازی و اپی‏تلیوم طبیعی مخاط دهان به روش ایمونوهیستوشیمی بود. مواد و روش‏ها: در این مطالعه مقطعی تظاهر P53 و PCNA در 53 نمونه از بلوک‏های پارافینی مربوط به لکوپلاکیا (بدون دیسپلازی (17 مورد)، دیسپلازی خفیف (15 مورد)، دیسپلازی متوسط (14 مورد) و دیسپلازی شدید (7 مورد) و 10 مورد مربوط به مخاط  طبیعی دهان به روش ایمونوهیستوشیمی بررسی شد. آزمون‏های آماری مورد استفاده آنالیز واریانس یک طرفه، کروسکال-والیس و ضریب همبستگی اسپیرمن بود. یافته‏ها: در این مطالعه، 40% از نمونه‏های نرمال، 5/76% از لکوپلاکیاهای بدون دیسپلازی و 33/93% از لکوپلاکیاهای دیسپلاستیک، P53 را بروز دادند و PCNA در تمامی این ضایعات تظاهر یافت. اختلاف معنی‏داری از لحاظ تعداد سلول‏های P53 و PCNA مثبت و شدت تظاهر این مارکرها بین ضایعات وجود داشت (001/0P<). ارتباط مثبتی بین تظاهر P53 و PCNA مشاهده گردید. نتیجه گیری: افزایش بروز P53 و PCNA با درجه دیسپلازی لکوپلاکیای دهان ارتباط دارد و از این دو نشانگر می‏توان برای درجه بندی دقیق تر دیسپلازی، همچنین یافتن ضایعات لکوپلاکیای دیسپلاستیک استفاده نمود. همچنین، می‏توان از PCNA و P53 بعنوان مارکرهای پیش بینی کنندة تغییرات بدخیمی در ضایعات لکوپلاکیای دهانی استفاده نمود.}, keywords_fa = {}, url = {https://jmds.mums.ac.ir/article_1283.html}, eprint = {https://jmds.mums.ac.ir/article_1283_466b9c761bfe2a43399d03f934634089.pdf} }
